No info yet but it looks like an alien invasion or an old indigenous civilization ala War of the Worlds or the Day the Earth stood still.

It will be directed by Alex Graves whose notable work includes the pilot episode for both Fox's Fringe and Journeyman.
No premiere date has been given, but due to comments made by NBC's Angela Bromstad, the show is expected to fill the Heroes time slot, saying, "‘Day One’ is a big event and we're looking at that to come into the 'Heroes' spot," Bromstad told the Reporter. "It's right now being looked at as a 13-episode run -- something people could commit to and we could make a big splash with."
